目的探討脂聯素對氣道平滑肌細胞( AMSCs) 增殖的影響及相關機制的研究。方法體外培養大鼠氣道平滑肌細胞株; RT-PCR 法檢測ASMCs 脂聯素受體的表達; 不同濃度的脂聯素( 5、10、20、40 及80 μg/mL) 作用于體外培養的大鼠ASMCs, 孵育不同時間, MTT 法檢測吸光值, 計算細胞抑制率; 不同濃度脂聯素( 5、10、20 及40 μg/mL) 作用于氣道平滑肌細胞48 h 后提取蛋白,Western blot 法檢測一磷酸腺苷活化蛋白激酶( AMPK) 及磷酸化一磷酸腺苷活化蛋白激酶( pho-AMPK) 的表達。結果 5 ～80 μg/mL 脂聯素在不同時間( 1、12、24、48 及72 h) 明顯抑制ASMCs 增殖, 并存在濃度依賴性( r =0. 324, P lt;0. 01) 和時間依賴性( r =0. 607, P lt;0. 01) ; Western blot 法檢測顯示空白對照組無pho-AMPK表達, 干預組隨著脂聯素濃度的升高AMPK 的活性增強( r =0. 907, P lt;0. 01) , pho-AMPK/AMPK 比值分別為( 27. 66 ±1. 03) %、( 31.91 ±0. 86) % 、( 75. 52 ±2. 67) % 和( 84. 50 ±1. 05) %, 差異有統計學意義( P lt;0. 05) 。結論脂聯素可能通過激活AMPK來抑制體外培養的ASMC 增殖。
